Joel (Jim Carrey) bliver forbløffet over at opdage, at hans kæreste Clementine (Kate Winslet) har fået deres kaotiske forhold slettet fra sin hukommelse. I ren og skær desperation kontakter han opfinderen af processen dr. Howard Mierzwiak (Tom Wilkinson) for at få den samme behandling. Men da minderne om Clementine begynder at fortage sig, går det pludselig op for Joel, hvor højt han stadig elsker hende.
